New Publication: Poly(allylamine)-tripolyphosphate Ionic Assemblies as Nanocarriers: Friend or Foe?
ABSTRACT: Designingeffectivedrugnanocarriersthat are easyto synthesize,robust,and nontoxicis a significantchallengeinnanomedicine.Polyamine-multivalentmoleculenanocomplexesare promisingdrugcarriersdue to theirsimpleand all-aqueousmanufacturingprocess.However,thesesystemscan presentissuesof colloidalinstabilityovertimeand cellulartoxicitydue to thecationicpolymer.In this study,we finelymodulatethe formationparametersof poly(allylamine-tripolyphosphate)complexestojointlyoptimizethe robustnessand safety.Polyallylaminewasionicallyassembledwithtripolyphosphateanionsto formliquid-like nanocomplexeswitha size of around200 nm and a zetapotentialof−30mV.We foundthatnanocomplexesexhibittremendouslong-termstability(9 monthsof storage)in colloidaldispersionandthattheyare suitableas protein-loadingagents.Moreover,the formationof nanocomplexesinducedbytripolyphosphateanionsproducesa switch-offin the toxicityof the systemby alteringthe overallchargefrompositiveto negative.Inaddition,we demonstratethat nanocomplexescan be internalizedby bone-marrow-derivedmacrophagecells.Altogether,thesenanocomplexeshaveattractiveand promisingpropertiesas deliverynanoplatformsfor potentialtherapiesbasedon the immunesystemactivation.
